The Engineer-it Kit

 

I received "The Engineer-it Kit" from Amino Labs. It is a bacterial transformation kit.

I read about this kit in the book I talked about in an earlier post - "Zero to Genetic Engineering Hero: The Beginner's Guide To Programming Bacteria At Home, School and In the Makerspace."

If you are a beginner in bio-engineering this is a good place to start. The kit is well priced, I felt shipping was a bit high but that is probably out of their control. It comes with almost everything you need, except the hardware. You do need a incubator (they do supply you with a video link to build a very simple one ), a hot and a cold water bath. I used an ice bath for my cold bath. They do have an all in one hardware that will do all of this, that you can purchase. Though it is expensive.
 
 

 
 
They made this kit to be as simple and easy to use as possible, even a beginner can do it. They supply you with everything you need:

Bacteria, plasmid, antibiotic, agar, petri-dishes, inoculation loop, streak template and even the sterile water.

They give you a manual.

Here is a video showing you all steps in the protocol.

You can even use a simulator to do the protocol.

If you are interested in bio-engineering this is a good way to start. As well they have other kits that you can purchase separately. This way it is not a huge outlay of money all at once.

 Second-Hand Store Finds

Second-hand stores can be a great place to find lab equipment and science kits. Though it is a hit or miss. Many times there is nothing to be had there. The biggest drawback can be the cost of the items. Some places charge outrageous prices for items. So be careful when looking and buying.

I found some electronic equipment I intend to use in some of my experiments.

1) A SOAR Corp. analog CRT based oscilloscope. From the simple tests I have done with it, it seems to be working fine.

2) A Cenco A.C. volt meter.

3) A electronic breadboard. To this I had to add the binding posts myself. Luckily I had some in my lab. The posts did not fit the holes. I did not want to buy any new ones so I designed and 3D printed fittings for the holes that the posts fit into. You can see the yellow fittings under the posts.

For less than $30 I picked up some useful equipment for my lab.

Another Science Kit

I found this kit at a second-hand store.

Magnetism Kit


"Magnetics Science Kit" - The science of magnets and ferrofluid. This second kit I also found for $10 at a second-hand store. I have wanted to start to look at ferrofluids and when I found this I figured this would be good way to start.
 
 




It comes with:
1 bottle of ferrofluid
iron filings
15 magnets
steel balls
copper tube
PVC track
Experiment guide
Etc.

I will more than likely still produce my own ferrofluids (you can find this online) but this gives me a start.

Finding kits at reduced or discounted prices (look after Christmas) and or used kits at second hand shops you are still able to get started in many different sciences and build your own lab.

I came across this kit, one at a department store.

The first kit "Charge" - This was on a clearance sale for $10 from $34.95. It was new in package. Taking a look at it and finding out it had the parts to build your own Van de Graaff generator, I figured I would like to build one so why not try it.

It's manual is very basic but a good start. From common items they show you how to build a Leyden Jar, Electroscope, electrostatic motor, Franklin Bell and of course the Van de Graaff generator. There are many youtube videos showing you how to build these items but for only $10 I get many of the pieces to build the generator.

I have been wanting to begin to experiment with high voltages and with this kit I should be able to start.

Books New and Old - Build It Yourself Science Laboratory

Written in 1963

This is one book that I found at my library when I was younger. It was one of a handful of books that taught me science. I would take it out as much as I was allowed to. I read and reread it over and over again. I created many of the projects and did many of the experiments that you find in the question sections.

I was already building my lab from what ever I could find and re-purpose for use at home. So I was so excited when I found this book. I believe I really wore out this book. It was still on the library shelves until around 8 or so years ago. One day I went to find it and it was no longer there. I did end up finding a used copy later on so I purchased it for my own library.

It is broken up into 3 sections: Chemistry, Physics and Biology. Physics was broken up into 6 other sections: Astronomy and Light, Atomic Energy, Electricity and Magnetism, Forces, Measurement and Motion, Geology and Meteorology.



Written 2015

The book has been re-edited and reprinted "the annotated Build-It-Yourself Science Laboratory" and now brings it up to date. Some things that are used in the original book are not legal or are dangerous or can not be found any longer. This new book helps to make it more useful for today's scientist.

Projects include:
Cloud Chamber
Vacuum pump
Microscope
Barometer
Radiometer
Microtome
Etc.

Over 100 pieces of equipment and 100's of projects. This book is a great way to learn to look at items and see other uses for them.

The Amateur Scientist, Procedures in Experimental Physics and Pictorial Handbook of Technical Devices

I love to read and I love to read scientific and tech books. Here are 3 that I had or received as gifts.

The Amateur Scientist - is one I had when I was much younger. As you can see it is quite worn. I have read this book cover to cover countless times. I have done many of the experiments in this book. This book is more of a how to do of experiments written in 1960 and is named for the column that the experiments are derived from - Scientific American's The Amateur Scientist monthly column, which ran for many years or decades really.

There are experiments in astronomy, physics, nuclear physics, rocketry, geology, math, biology, optics, etc. It is so full of different experiments it is amazing. Many of the experiments are dangerous and maybe hard to do as many items have become very hard to find. It is a great read even if you can't do the experiments. It does show what individuals can do and accomplish on their own. You maybe able to find this book as a pdf file on line.

The next 2 books were given to me as gifts over the last few years. Both as well are older than most.

Procedures in Experimental Physics - was written in 1938 and it covers many procedures needed in a laboratory, such as, glass blowing, optical work, vacuums, surface coating, electronics, old time photography, etc. Another great read in how build and use science equipment. A bit out-dated but a great read.

Pictorial Handbook of Technical Devices - written in 1971, this book shows you pictures of many many devices so you can have an understanding of how they are put together and how they work. There are joints, springs, levers, belt drives, prisms and lenses, etc. It is full of devices.

These books are out of print but you may be able to find them online. Even though they are older books they allow me to see how things can work and helps me come up with new ideas for my own experiments. I will be discussing more books in the near future. I may even start a book of the month column. In the mean time, old or new keep reading anything that interests you.

Sterile Chamber

I found this item, a baby incubator cover that was very inexpensive. I am using it to build a sterile chamber to do culturing in. It has plenty of room inside it and plenty of arm maneuverability to be comfortable to do projects in it.

I will mount it on a vinyl covered piece of plywood or press board. Add a fan that blows the air out and in through a Hepa filter. I will sterilize it with a Lysol/alcohol solution. This will keep the work area in a fairly stable environment, I won't be breathing over the work area or as I move sending dust and particles into the chamber. This should work better than the aquarium that was put together earlier.

I will keep post further as I finish it up.

Even More Books

Older Books

I have picked up 3 more books for my library. 2 are older and from a better time for citizen science. The other one newer but important for the amateur scientist. (I truly dislike using amateur or citizen scientist. It gives the impression that real science can not be done by them.)

1) Apparatus - 1967 by F. F. Blackwell.

This book shows you how to build various science equipment from household materials. Focusing more on the physical sciences but can be applied to the biological as well. Sections include:

Elementary electricity, Biological apparatus, Light, Machines, Air, Sound and heat, Water, Weather and soil studies.

2) Procedures in Experimental Physics - by John Strong. Originally published in 1938 republished 1986.

Another book dealing with physics. It has great explanations of certain techniques. This book and The Amateur Scientist by C.L. Stong make for great physics experiments of old. Sections include:

Glass blowing, High Vacuum technique, Optical work, Photoelectric cells, Heat and high temperature, Molds and casting, etc.

3) The Laboratory Companion: A Practical Guide to Materials Equipment and Technique - by Gary S. Coyne 2006.

This is a very useful book that discusses the different materials used in labs and how to maintain them. This is a modern book discussing modern equipment.

When I look at some of the older books I pick up I can't believe how many books existed that helped young people build their own labs and equipment. A far cry from what you find today. I am glad that I am still able to find some of them.

Another Old Science Kit



Another Old Science Kit

Here in the top picture you see another set I found, it doesn't have the box though. This is exactly the one my parents bought for me in the 1970's. It is the "Mr. Wizard Experiments in Crystal Growing". I really enjoyed playing with this, I always liked growing crystals. As you can see they gave you plenty of chemicals to work with back then. Mr. Wizard had some nice science kits in the 1970's. I had 2 of them the chemistry set and the crystal growing set.

The second picture is an old catalog from the The Perfect Parts Company. I remember once when we I was young and we where in the United States. I don't remember what store it was but they had a booth with this companies products and there was a huge line up of people trying to buy things. I had wished we had some place like this where I lived, finding lab equipment at a reasonable cost was hard for me when I was young. If you do web check you will find they still exist.

DIYBIO and Genetics

Over the past weekend I did some web surfing and I found this site called "Learn Genetics":

It teaches the basics of genetics. It shows you how to virtually build a DNA molecule, talks about DNA to protein and all the basics needed to understand genetics. There virtual labs such as 1) DNA extractions, 2) PCR, 3) Gel Electrophoresis and 4) DNA Microarray.

A lot of what is on there can be transferred to using plastic dishes instead of building the chamber completely form scratch. But still the lessons for working with plastics are good an can be beneficial if you go that route.

Books

It has been awhile since I added any information. I have spent some time in building my incubator. Testing it out exactly. I will be placing the fish tank heater into a glass dish of water to heat the incubator. I tested this out and it seems to work quite well. I will add either some copper sulphate or aquarium anti-microbial solution to the water to keep microbial life from growing in the glass dish. I need to come up with some shelves to hold plates, tubes and flasks at different heights in the incubator. I will post a final picture of the incubator when it is finished.

For father's day I received a wonderful book titled: Build-it-Yourself Science Laboratory by Raymond E. Barrett. It was published in 1963. It shows you how to build all kinds of lab equipment, for chemistry, physics, and biology. It even has questions to answer by scientific experimentation after each section. We need a book like this today for building our own labs.

Other books I have picked up for my growing library:

Magic with Chemistry by Edward L. Palder 1964.

150 Science Experiments Step-By-Step by Judith Viorst 1963.

Physics, Fun and Beyond by Eduardo de Campos Valadares 2006.

The Thomas Edison book of Easy and Incredible Experiments by the Thomas Alva Edison Foundation 1988.

Basic Chemistry Experiments: A Golden Hobby Book by Robert Brent 1965 edition.

So now I have many more books to add to my collection and more experiments to try out.
